As Damon Albarn, Alex James, Graham Coxon and Dave Rowntree made their way up Fort Hill in Margate, they were mobbed by fans, who had travelled from as far as Denmark, France and Northern Ireland.
They ate fish and chips from Pete's Fish Factory in The Parade, before the gig kicked off just before 9pm.
The band's two-hour set included their hits Girls And Boys, Parklife, Country House and Song 2.
The gig was the first of four warm-up shows ahead of their sold-out concert at London's Hyde Park on August 12.
Damon told the Winter Gardens audience: "Thank you for bearing with us tonight. We love you very much."
Fans roared with appreciation as the band returned to the stage for an encore, which concluded with The Universal from album The Great Escape.
Dedicated giggers camped out overnight to bag themselves tickets, which sold out in 45 minutes.
Television presenter Holly Willoughby was at the gig. Before the boys took to the stage she tweeted: "Damon, Graham, Alex and Dave....let's go. #margate."
She also uploaded a photograph of herself in the excited crowd and tweeted: "So I'm playing yeah!"
Earlier this year, Blur won a lifetime achievement award at the Brit Awards.
They will go on to play in Wolverhampton, Plymouth, festivals in Denmark and Sweden, before playing their sold-out Olympic show with The Specials and New Order at Hyde Park on August 12.
